Abstract:Objective To investigate the expression of T cell immunoglobulin domain and mucin domain protein-3 (Tim-3), galectin-9 (Gal-9) and interferon-γ (IFN-γ) in oral lichen planusand (OLP) tissues and the role of Tim-3/ Gal-9 pathway in the formation of OLP and its significance. Methods From October 2016 to October 2019, 41 cases of OLP patients treated in the Department of Periodontal Mucosa, Stomatological Hospital of Guizhou Medical University (“our hospital” for short) were selected as the OLP group, and 10 normal oral mucosal people treated in our hospital during the same period were selected as the normal group. The expression of Tim-3, Gal-9, and IFN-γ in the two groups and their correlation were detected by immunohistochemical SABC method, at the same time, the correlation between the pathological characteristics of OLP patients and Tim-3, Gal-9 and IFN-γ were analyzed. Results The positive expression rates of Tim-3 and IFN-γ in the OLP group were higher than those in the normal group, and the positive expression rates of Gal-9 was lower than that in the normal group, with statistical significance (P < 0.05). In OLP tissue, the expression of Tim-3 was positively correlated with the epithelium and lamina propria (r > 0, P < 0.05); the expression of Tim-3 and Gal-9 was negatively correlated in the epidermis (r < 0, P < 0.05); and the expression of Tim-3 and IFN-γ was positively correlated in lamina propria (r > 0, P < 0.05). The score of lymphocyte infiltration was positively correlated with the positive expression of Tim-3 in the epithelium and lamina propria (r > 0, P < 0.05), and negatively correlated with Gal-9 (r < 0, P < 0.05); the score of liquefaction infiltration of basal cells was positively correlated with the positive expression of Tim-3 and IFN-γ in the epidermis (r > 0, P < 0.05). Conclusion The high expression of Tim-3 and low expression of Gal-9 in the lesions of OLP patients may be due to the abnormal Tim-3/Gal-9 pathway, which enhances T helper cell 1 effector cells and promotes the secretion of IFN-γ, and is involved in the process of pathological injury in the lesion area of OLP.
