Effects of Jianpi Yiqi Formula on gastric tissue of rats with chronic atrophic gastritis with syndrome of spleen deficiency

Abstract:Objective To observe the effect of Jianpi Yiqi Formula on gastric tissue of rats with chronic atrophic gastritis (CAG) with syndrome of spleen deficiency. Methods Fifty-five healthy SD rats were selected to make the model of rat with CAG with syndrome of spleen deficiency by MNNG + Ranitidine method. After 24 weeks of intervention, five of them were selected to observe the morphology of their gastric mucosa to confirm the success of modeling. The last rats were divided into Jianpi Yiqi Formula low (13.1 g/ [kg·d]), medium (26.2 g/ [kg·d]) and high dose group (52.4 g/ [kg·d]), and model group (normal saline), positive drug group (folic acid 2.7 mg/ [kg·d] + Teprenone 13.5 mg/ [kg·d]) by random number table method, with ten rats in each group, another ten healthy SD rats were selected as blank group (normal saline). The status and body weight of the six groups were recorded by gavage for eight weeks. The pathological changes of the gastric tissues were observed by HE method. The changes of serum gastrin and somatostatin (SST) levels were determined by enzyme linked immunosorbent assay, and the protein expressions of Ki67, Notch1, Notch2 and Hes1 were determined by immunohistochemical stain. Results Blank group fur was white and shiny, active. The fur of the model group was yellow and easy to fall off, and the activity was reduced. The positive drug group and the low-dose, medium-dose and high-dose groups of invigorating spleen and replenishing qi formula had bright fur and high activity. The body weight of the model group was lower than that of the blank group, and the body weight of the Jianpi Yiqi Formula medium-dose and high-dose groups were higher than that of the model group, with statistical significance (P < 0.05 or P < 0.01). The results of HE staining showed that the gastric mucosa epithelial layer was clear and the glands were normal in the blank group. Gastric mucosal epithelium in model group was thinned, glands were reduced, arranged loosely and disorderly, and goblet cells could be seen. In the other four groups, the gastric mucosal epithelium was thickened, the glands were increased, the arrangement tended to be orderly, and the infiltration of inflammatory cells was decreased. Serum gastrin and SST levels in model group were lower than those in blank group, while the levels of serum gastrin in Jianpi Yiqi Formula high-dose group was higher than that in model group, and the levels of SST in Jianpi Yiqi Formula low-dose, medium-dose and high-dose groups were higher than that in model group, with statistical significance (P < 0.05 or P < 0.01). The protein expressions of Ki67, Notch1, Notch2 and Hes1 in model group were lower than those in blank group, the protein expression level of Ki67 in Jianpi Yiqi Formula low-dose group was higher than that in the model group, the protein expression levels of Notch1 and Notch2 in Jianpi Yiqi Formula low-dose, medium-dose and high-dose groups were higher than that in the model group, and the protein expression level of Hes1 in Jianpi Yiqi Formula high-dose group was higher than that in the model group, with statistical significance (P < 0.05 or P < 0.01). Conclusion Jianpi Yiqi Formula can improve the pathological state of gastric mucosa atrophy in CAG rats with syndrome of spleen deficiency, and the mechanism is related to the up-regulation of the expression of proteins related to Notch and other signaling pathways.
