Study on protective effects and mechanism of liquiritin on photoaging hacat cells induced by UVB

Abstract:Objective To investigate the protective effects and the mechanism of liquiritin on photoaging human keratinocytes (HaCaT) cells induced by ultraviolet B (UVB). Methods The different concentrations of liquiritin were used on HaCaT cells, the best effective and safe concentration of liquiritin was determined by methyl thiazolyl tetrazolium(MTT). The photoaging model was set up by the irradiation of UVB at irradiation dose of 30 mJ/cm2. It was used to treat liquiritin + UVB group cells 24 h with 10-7, 10-6, 10-5 mol/L, the effects of liquiritin on the proliferation rate of photoaging model HaCaT cells were detect by MTT. The activity of glutathione (GSH), catalase (CAT) and the content of malondialdehyde (MDA) were detected by the corresponding kits. The detection of interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) protein expression were detected by Western blot. Results The liquiritin with the concentrations of 10-7, 10-6, 10-5 mol/L had no obvious effect on the proliferation and inhibition of HaCaT cells (P > 0.05), which were the best safe and effective concentrations. Compared with the blank group, the cell proliferation rate of the UVB group was significantly decreased, the activity of GSH and CAT was significantly decreased, and the content of MDA was significantly increased, the expression of IL-1β, IL-6 and TNF-α protein were increased (P < 0.01). Compared with the UVB group, the cell proliferation rates of the (10-7, 10-6, 10-5) mol/L liquiritin+UVB group were significantly increased, the activity of GSH and CAT was significantly increased, the content of MDA was significantly decreased, the expression of IL-1β, IL-6 and TNF-α protein were significantly decreased (P < 0.05 or P < 0.01). Conclusion Liquiritin can protect HaCaT cells photoaging, which mechanism is related to the improvement of antioxidant enzyme activity and inhibition of inflammatory cytokines.
