Optimization of extraction process of Qinkou Huashi Mixture based on entropy weight method combined with Box-Behnken design-response surface methodology

Abstract:Objective To optimize the extraction process parameters of Qinkou Huashi Mixture based on entropy weight method combined with Box-Behnken design-response surface methodology. Methods Using high performance liquid chromatography and ultraviolet-visible spectrophotometry, the amount of baicalin, total flavonoids, and total solids were used as evaluation indexes, extraction times, extraction time, and water addition multiple were used as investigation factors. The weight coefficients of each index were analyzed by entropy weight method, and Box-Behnken design-response surface method was used to optimize the optimum extraction parameters of Qinkou Huashi Mixture. Results The contents of baicalin and total flavonoids showed a good linear relationship in the range of 6.06-193.68 and 1.96-31.25 μg/ml (r=0.999 8, 0.999 5). The weight coefficients Wj of baicalin, total flavonoids, and total solids were 0.467 9, 0.287 1, and 0.245 0. The optimal extraction process of Qinkou Huashi Mixture was determined as follows: extraction times were 2 times, extraction 33 min and each time was addition 7.8-fold water. Conclusion The optimized extraction process is stable, reliable, and reproducible, which can be used for mass production.
