Study on the effect and mechanism of senile functional diarrhea treated by Sishen Pills based on brain-gut axis

Abstract:Objective To observe and analyze the effects of Sishen Pills on the contents of 5-hydroxytryptamine (5-HT), substance P (SP), and glutathione peroxidase (GSH-Px) in senile functional diarrhea rats. Methods Forty SPF 12-month- old male SD rats with body weight (380±20) g were divided into blank group, model group, positive drug group, and Sishen Pills group by random number table method, with ten rats in each group. The rats were injected with hydrocortisone (25 mg/kg) and gavaged with senna leaf (0.45 g/ml) to create models. Body weight, body temperature, diarrhea index, urinary D-xylose excretion rate of rats were recorded. During administration period, the Sishen Pills group and positive drug group were given 0.183 g/ml Sishen Pills suspension, 1.523 mg/ml Piveronium Bromide suspension, respectively. The blank group and model group were given equal volume of normal saline. Continuous administration for 14 days. The contents of 5-HT, SP, and GSH-Px in hypothalamus and colon were observed. Results In Sishen Pills group, one died due to improper operation in the second week of modeling; in model group, one died due to stomach cold in the fourth week of modeling. There were nine rats in each Sishen Pills group and model group. Before and after administration, compared with blank group, body weight and body temperature were decreased, diarrhea index was increased, urinary D-xylose excretion rate was decreased in model group, and the differences were highly statistically significant (P<0.01). After administration, compared with model group, body weight and body temperature were increased, diarrhea index was decreased, urinary D-xylose excretion rate was increased in Sishen Pills group, the differences were highly statistically significant (P<0.01). Compared with blank group, the contents of 5-HT and SP in model group were increased, while the content of GSH-Px was decreased, and the differences were highly statistically significant (P<0.01). Compared with model group, the contents of 5-HT and SP in Sishen Pills group were decreased, while the content of GSH-Px was increased, and the differences were highly statistically significant (P<0.01). Conclusion Sishen Pills can effectively treat senile functional diarrhea of spleen-kidney-yang deficiency, its mechanism may be related to the regulation of the content and expression of 5-HT, SP, GSH-Px of brain-gut peptides in brain-gut axis.
