Abstract:Objective To investigate the effect of Tangluoning regulating miR-211 on apoptosis of dorsal root ganglion neurons (DRGn) in diabetic peripheral neuropathy rats. Methods Neonatal rat DRGn cells were used as research objects.They were divided into normal group, high glucose group, Tangluoning group, miR-211 inhibitor negative group, and miR-211 inhibitor group. The normal group was treated with special medium for neurons, while the high glucose group, the Tangluoning group, the miR-211 inhibitor negative group, and the miR-211 inhibitor group were treated with special medium for high glucose neurons (containing 50 mmol/L glucose). miR-211 inhibitor negative group and miR-211 inhibitor group were transfected with empty vector and miR-211 inhibitor vector, respectively. The normal group, the high glucose group, the miR-211 inhibitor negative group and the miR-211 inhibitor group were given 10% blank serum intervention, and the Tangluoning group was given 10% Tangluoning containing serum intervention. After 24 hours of intervention, the cells in each group were collected to detect the apoptosis rate, miR-211, and the expression levels of apoptosis pathway related protein kinase R-like endoplasmic reticulum kinase (PERK), eukaryotic initiation factor 2α (eIF2α), activator of transcription 4 (ATF4), and C/EBP homologous protein (CHOP). Results There was no significant difference in every indexes between high glucose group and miR-211 inhibitor negative group (P > 0.05). The apoptosis rate, miR-211 gene expression, and protein expression levels of PERK, eIF2α, ATF4, and CHOP in the high glucose group were higher than those in the normal group, the apoptosis rate, miR-211 gene expression, and protein expression levels of PERK, eIF2α, ATF4, and CHOP in Tangluoning group and miR-211 inhibitor group were lower than those in high glucose group, and the differences were statistically significant (P < 0.05). Conclusion Tangluoning can attenuate DRGn apoptosis induced by high glucose by inhibiting miR-211 regulating apoptosis pathway.
