Animal experimental study of Yinxieping in the treatment of psoriasis vulgaris

Abstract:Objective To establish animal models of Balb/c nude mice with psoriasis vulgaris, observe the effect of Yinxieping on double adjustable epidermal tissue protein (AREG), vascular endothelial growth factor (VEGF) and keratinocyte growth factor (KGF). Methods 36 Balb/c nude mice were randomly divided into model control group and Yinxieping A group, Yinxieping B group, the three groups were all smeared imiquimod cream for consecutive 8 d in skin of 4 sections in head and neck, upper limbs, torso and lower extremities of mice to built the model. At the dose of 8th day after modeling, Yinxieping A group, Yinxieping B group were treated by gavage for 10 d with 0.1 g/kg and 0.5 g/kg dose, model group was injected into the same amounts saline. At the treatments 8th, 10th day, psoriasis area and severity index (PASI) was used to evaluated the animal head neck, upper limbs, torso and lower extremities 4 skin lesions; ELISA was uesd to determine contents of AREG, KGF, VEGF and vascular endothelial growth factor receptor 1 (VEGFR1) and other indicators in epidermal tissue homogenates in a fixed quantity. Results PASI scores of Yinxieping A group, Yinxieping B group and model control group at modeled 8th day were compared, the differences were not statistically significant (P > 0.05), when treated by gavage at 10th day, compared with the model control group and themsavies at modeled 8th day, PASI scores of Yinxieping A group, Yinxieping B group decreased significantly (P < 0.05), the expression of AREG, KGF, VEGF and VEGFR1 in epidermal tissue homogenates was lower (P > 0.05). Yinxieping A group and Yinxieping B group were compared, the differences were not statistically significant (P > 0.05). Conclusion Yinxieping can decrease contents of AREG, KGF, VEGF in epidermal tissue homogenate, and it may block the combination of VEGF and VEGFR1, which has positive effects in the inhibition of epidermal keratinocyte lesions and subcutaneous tissue angiogenesis, reducing the subjects change in animal with psoriasis to play parts in pathological intervention.
