The value of Xuebijing Injection on the rehabilitation of rabbits with acute lung injury induced by firearm injury and the effect on the expression of NF-κB in lung tissue

Abstract:Objective To investigate the value of Xuebijing Injection (XBJ) on the rehabilitation of rabbits with acute lung injury (ALI) induced by firearm injury and the effect on the expression of nuclear factor kappa B (NF-κB) in lung tissue. Methods Sixty healthy male rabbits were randomly divided into normal control group, ALI model group, normal saline group, low dose of XBJ treatment group, medium dose of XBJ treatment group, high dose of XBJ treatment group by random number table mehtod, with 10 rats in each group. The rabbits of ALI model group, normal saline group, low dose of XBJ treatment group, medium dose of XBJ treatment group, high dose of XBJ treatment group were inhaled with high-temperature gas, so as to complete the model of rabbits with ALI. After modeling for half an hour, low, medium, high dose of XBJ treatment groups were respectively received intraperitoneal injection of 10, 20, 30 mg/kg XBJ, normal saline group was injected with normal saline, while normal control group and ALI model group were not dealt with any treatment. After 12 h, the rabbits were killed and the lung tissue of left upper lobe was taken to detect the expression of NF-κB in lung tissue; under the light microscope, the lung tissue was taken to make pathological examination, and the pathological changes were recorded in time. Results The results of pathological examination: in the normal control group, the alveolar structure was clear, the alveolar wall was thin, and no edematous fluid was found in the lung tissue; in the ALI model group and normal saline group, the alveolar structure was fuzzy, the alveolar wall was broken, there were a lot of inflammatory cells infiltration in the lung, obvious edema was found in the pulmonary mesenchyme and alveolar space, many red blood cells could be seen, and the mesenchyme of alveolar wall was thickened, the hyaline membrane was formed; after treatment by XBJ, the alveolar structures in all groups were still existed, the inflammatory cells infiltration in the lung was reduced, pulmonary mesenchyme and alveolar space showed moderate edema, less cataclysm and bleeding could be seen. Western blot of lung tissue showed that, the activity of NF-κB in ALI model group was significantly higher than that of normal control group (P < 0.01); compared with ALI model group and normal saline group, the activity of NF-κB in lung tissue of low, medium, high dose of XBJ treatment groups were all decreased, the differences were all highly statistically significant (P < 0.01), among which, medium dose of XBJ treatment group was decreased most significantly, which had statistically significant differences compared with those of low, high dose of XBJ treatment groups (P < 0.05, P < 0.01). Conclusion XBJ injection has protective effects on rabbits with ALI induced by firearm injury, and the protective effect of medium dose of XBJ is the most obvious.
