Abstract:Objective To explore the role and mechanism of long noncoding RNA-ANRIL (lncRNA ANRIL) in proliferation, invasion and apoptosis of breast cancer cells. Methods Breast cancer cells were transiently transfected with si-ANRIL. Real-time PCR was used to confirm the interfere effect of si-ANRIL, MTT assay was used to detect cell proliferation, transwell method was used to detect cell invasion, flow cytometer (FCM) was used to detect cell apoptosis, Western blot was used to detect the protein expression level of p53. Breast cancer cells were co-transfected with si-ANRIL and si-p53, MTT assay, transwell method and FCM were used to detect cell proliferation, invasion and apoptosis. Results ANRIL knockdown decreased cell proliferation, invasion, and increased cell apoptosis (P < 0.05). ANRIL knockdown activated p53 signal pathway. Simultaneous p53 knockdown partially reversed the effect of ANRIL knockdown. Conclusion LncRNA ANRIL can promote the proliferation, invasion and inhibit apoptosis of breast cancer cells by inhibiting p53 pathway, thus promoting the occurrence and development of breast cancer.
