Effect of three knee stitches on the morphological and behavioral changes of knee osteoarthritis model rat cartilage

Abstract:Objective To investigate the effect of three knee stitches on the morphological and behavioral changes of knee osteoarthritis (KOA) model rats. Methods A total of 28 male SPF SD rats (three to four weeks old, weight 180 to 220 g) were divided into blank group and model group with eight rats, acupuncture group with six rats, and western medicine group with six rats respectively by random number table method. The rat model of KOA was established by anterior cruciate ligament transection except the blank group. After five weeks of modeling, the acupuncture group received three knee stitches from the left knee joint for acupuncture treatment, once a day, and rested for five consecutive days for two days for three weeks. In the western medicine group, Diclofenac Diethylamine Latex was applied outside the affected knee. Blank group and model group were not involved. The morphological changes of cartilage after X-ray film, Lequesne MG score, Mankin’s score, cartilage, and HE staining were observed and compared. Results Compared with blank group, the left knee articular surface space narrowed, articular surface wear, and osteophyte formation were obvious in model group, articular cartilage surface defect was obvious, the number and distribution of cells were decreased, and Lequesne MG and Mankin’s scores were increased (P<0.05). Compared with model group, Lequesne MG and Mankin’s scores in acupuncture group were decreased (P<0.05), the surface of cartilage was slightly rough and shiny, and the number of cells was increased and arranged in a clear and orderly manner. The Lequesne MG scores in model group, acupuncture group and western medicine group after treatment were lower than those before treatment (P<0.05). Conclusion Three knee stitches can improve the morphologic and behavioral indexes of the cartilage and protect the knee joint of KOA rats.
