Effect of local injection of hepatocyte growth factor transfected bone marrow mesenchymal stem cells on tendon-bone healing after anterior cruciate ligament reconstruction in rabbits

Abstract:Objective To explore the effect of hepatocyte growth factor (HGF) transfected bone marrow mesenchymal stem cell (MSC) on tendon-bone healing after anterior cruciate ligament (ACL) reconstruction in rabbits. Methods Forty-five male New Zealand rabbits (3.0-3.5 kg) aged three to five months were divided into groups A, B, and C by random number table method, with 15 rabbits in each group. All three groups underwent double hindlimb ACL reconstruction, group A did not inject any substance after ACL operation; in group B, MSCs fibrin glue was injected into the tibial canal after operation; in group C, HGF gene modified MSCs fibrin glue was injected into the tibial canal after operation. Six weeks after operation, HE staining was used to observe the tendon-bone healing; the maximum tensile load and the maximum stretching distance of the graft tendon exiting from the tibial canal were tested; the expression of fibroblast growth factor 2 (FGF2) mRNA and SMAD4 mRNA in bone canal and HGF mRNA in joint fluid were detected by RT-PCR. Results Six weeks after operation, the tendon-bone interface of group A was loosely connected and collagen fibers were disordered; in group B, the tendon-bone interface was closely connected and the gap was small, but the arrangement of collagen fibers was not orderly; in group C, the tendon-bone interface was closely connected and the collagen fibers were neatly arranged. The maximum tensile load of group B was higher than that of group A, and that of group C was higher than that of group B (P<0.05); there was no significant difference in the maximum stretching distance between groups A, B and groups B, C (P>0.05). The FGF2 mRNA and SMAD4 mRNA in bone canal in group B were higher than those in group A, and those in group C were higher than those in group B (P<0.05); HGF mRNA of joint fluid in group C was higher than that in group B (P<0.05). Conclusion Local injection of HGF transfected bone marrow MSCs can enhance tendon-bone healing after ACL reconstruction in rabbits.
