参芎葡萄糖注射液对低灌注大鼠血-脑脊液屏障通透性的影响

摘要 目的 研究参芎葡萄糖注射液对低灌注大鼠血-脑脊液屏障通透性的影响。 方法 60只SPF级SD雄性大鼠采用随机数字表法分为假手术组、低灌注组和参芎组,每组20只。低灌注组和参芎组结扎颈总动脉制作脑缺血动物模型,假手术组只暴露颈总动脉不结扎。参芎组腹腔注射参芎葡萄糖注射液24 mg/kg,假手术组和低灌注组腹腔注射等量生理盐水。酶联免疫吸附试验检测血清白蛋白(SAlb)和脑脊液白蛋白(CAlb)含量;实时荧光定量PCR检测脉络丛组织ZO-1与Occludin的mRNA;激光共聚焦方法检测脉络丛组织ZO-1与Occludin的荧光强度。 结果 低灌注组CAlb、脑脊液及血清白蛋白商值(QAlb)高于假手术组,参芎组CAlb、QAlb低于低灌注组,差异有统计学意义(P < 0.05);低灌注组与假手术组SAlb比较,差异无统计学意义(P > 0.05);参芎组与低灌注组SAlb比较,差异无统计学意义(P > 0.05)。低灌注组ZO-1、Occludin mRNA、荧光强度水平低于假手术组,参芎组ZO-1、Occludin mRNA、荧光强度高于低灌注组,差异有统计学意义(P < 0.05)。 结论 低灌注状态下,大鼠的血-脑脊液屏障通透性增加,参芎葡萄糖注射液在一定程度上对血-脑脊液屏障有保护作用。

Abstract:Objective To study the effect of Shenxiong Glucose Injection on the permeability of blood-cerebrospinal fluid barrier in hypoperfusion rats. Methods Sixty SPF male SD rats were divided into sham operation group, hypoperfusion group and Shenxiong group by random number table method, with 20 rats in each group. In the hypoperfusion group and Shenxiong group, the common carotid artery was ligation to make an animal model of cerebral ischemia, while in the sham operation group, only the common carotid artery was exposed without ligation. Shenxiong group was intraperitoneally injected with Shenxiong Glucose Injection 24 mg/kg, sham operation group and hypoperfusion group were intraperitoneally injected with normal saline. Serum albumin (SAlb) and cerebrospinal fluid albumin (CAlb) were determined by enzyme linked immunosorbent assay. The mRNA of ZO-1 and Occludin in choroid plexus was detected by real-time fluorescence quantitative PCR. The fluorescence intensity of ZO-1 and Occludin in choroid plexus was detected by laser confocal method. Results CAlb and serum albumin and cerebrospinal fluid quotient (QAlb) in the hypoperfusion group were higher than those in the sham operation group, and CAlb and QAlb in the Shenxiong group were lower than those in the hypoperfusion group, with statistical significance (P < 0.05). There was no significant difference in SAlb between the hypoperfusion group and sham operation group (P > 0.05). There was no significant difference in SAlb between the Shenxiong group and hypoperfusion group (P > 0.05). The mRNA and fluorescence intensity of ZO-1 and Occludin in the hypoperfusion group were lower than those in the sham operation group, and the mRNA and fluorescence intensity of ZO-1 and Occludin in the Shenxiong group were higher than those in the hypoperfusion group, with statistical significance (P < 0.05). Conclusion The permeability of blood-cerebrospinal fluid barrier increased in rats under hypoperfusion state, and Shenxiong Glucose Injection had protective effect on blood-cerebrospinal fluid barrier to some extent.
