Effects of jiájǐ point electroacupuncture on spontaneous pain behavior and hyperalgesia in central pain model rats after experimental spinal cord injury

Abstract:Objective To observe the effects of jiájǐ point electroacupuncture on spontaneous pain behavior and hyperalgesia in central pain model rats after experimental spinal cord injury (SCI). Methods Thirty SPF male SD rats (56 days of age, weight 230-240 g) were divided into blank control group, SCI group and jiájǐ point group by random number table method, with ten rats in each group. In the SCI group and the jiájǐ point group, Allen’s percussion method was used to establish the experimental SCI central pain model at the L1 segment of the spinal cord, and the blank control group was not operated to create the model. The jiájǐ point group was treated with direct acupuncture at T8-T12 jiájǐ point for 30 min, once a day, for 14 days, while the blank control group and the SCI group were only fixed (30 min). The spontaneous pain behavior of rats was observed during treatment. After 14 days of treatment, Spinal cord motor evoked potential (scMEP) was recorded by unipolar transcortical electrical stimulation. Somatosensory evoked potential (SEP) in L1 was recorded by stimulation. Mechanical pain threshold was determined by von Frey hair method. The hot plate method was used to determine the hyperalgesia. Basso Beattie Bresnahan (BBB) score and oblique plate test were used to evaluate the treatment effect. Results The incidence of spontaneous screaming, scratching, licking, biting and autophagy in the jiájǐ point group was lower than that in the SCI group, and the difference was statistically significant (P < 0.05). Compared with the SCI group, N1 and N2 peaks of early components of scMEP in the jiájǐ point group were increased, and the latency of SEP, P and N waves were shortened, the pain response threshold of the lower trunk of the injured plane to non-invasive mechanical light pressure and light touch and 50% forepaw lifting in the jiájǐ point group were increased, the Paw withdrawal latency of the jiájǐ point group was increased, the paw withdrawal latencies were prolonged, the incidence of hyperalgesia was decreased, the BBB score was increased, and the inclination of inclined plate was increased, with statistical significance (P < 0.05). Conclusion Electroacupuncture at jiájǐ point can improve the pain threshold, dredge the meridians of “governor channel injury” and improve the spontaneous pain and hyperalgesia, which is conducive to the recovery of nerve conduction and motor function.
