Correlation between expression of ASH1L and its related regulatory factors in peripheral blood of patients with oral lichen planus and immune function

Abstract:Objective To analyze the correlation between the expression of ASH1L and its related regulatory factors in the peripheral blood of patients with oral lichen planus (OLP) and the immune function, and to explore its role and significance in the pathogenesis of OLP. Methods The peripheral blood of 40 OLP patients (OLP group) and 20 healthy volunteers (control group) were collected from the Stomatological Hospital Affiliated to Guizhou Medical University from August 2018 to August 2019. Flow cytometry and scattering turbidimetry were used to detect the levels of cellular immunity and humoral immunity. The relative expression levels of ASH1L, ubiquitin editing enzyme A20, and interleukin-6 (IL-6) mRNA in peripheral blood mononuclear cells of the two groups and their correlation with OLP immune indexes were detected by quantitative real-time PCR. Results The levels of CD3+, CD3+CD4+, CD3+CD8+, total T+B+NK cells, and IgM in OLP group were lower than those in control group, and CD19+, CD4+/CD8+, complement C3, and C4 levels were lower than those in control group, with statistical significance (P < 0.05). The relative expression levels of ASH1L and A20 mRNA in OLP group were lower than those in control group, and IL-6 mRNA in OLP group was higher than that in control group, with statistical significance (P < 0.05). The relative expression of ASH1L mRNA in OLP group was positively correlated with the level of IgG (r > 0, P < 0.05). The relative expression of A20 mRNA was positively correlated with the levels of CD8+ and IgA (r > 0, P < 0.05), and negatively correlated with the CD4+/CD8+ (r < 0, P < 0.05). Conclusion The abnormal expression of ASH1L and its related regulatory factors in the peripheral blood of OLP patients is correlated with the level of immune indicators, and plays a certain role in the immune-related pathogenesis of OLP.
