Changes of early islet function in pigs with severe heat stroke

Abstract:Objective To observe the changes of early islet function in pigs with severe heat stroke. Methods Twelve healthy CV level male Bama miniature pigs were divided into control group (six pigs) and model group (six pigs) according to random number table method. The control group was placed at room temperature, and the model group was placed in the climate chamber with constant temperature and humidity. The severe heat stroke model of pigs was established under the conditions of 44-47 ℃ and relative humidity of (65±5) %. The core temperature of the model group > 42℃ was regarded as the success of modeling, that was the experiment ended. General conditions and core temperature of the two groups were monitored. Blood samples were collected from the two groups for examination after the end of the experiment to compare the changes in blood glucose and insulin levels. hematoxylin-exosin staining was used to observe the tissue morphology of the islets, liver and kidney of the two groups, and immunohistochemical staining was used to observe the expression level of insulin in the islets of the two groups. Results After the model group entered the constant temperature and humidity climate cabin, the core temperature of the model group showed an increasing trend, while the core temperature of the control group did not change. At the core temperature > 42℃, the model group showed depression of spirit, unstable gait, conjunctival congestion, and weakened or disappeared corneal reflex, while the control group showed no abnormal symptoms. There was no significant difference in blood glucose and insulin levels in the control group before and after the experiment (P > 0.05). After the experiment, the blood glucose level of the model group was higher than before, and the blood insulin level of the model group was lower than before, and the blood glucose level of the model group was higher than that of the control group, and the blood insulin level of the control group was lower than that of the control group, the difference was statistically significant (P < 0.05). After the experiment, there were no significant changes in islet, liver and kidney tissue in the control group. The islet margin of model group was not clear, the shape was irregular, the cell edema, a few granulocytes infiltration, and the blood stasis of the capillary. The renal tubules and collecting ducts were dilated, thrombosis was observed in the vessels, epithelial granules were degenerative, and a few inflammatory cells were infiltrated. The structure of hepatic lobules was disordered, the hepatic sinuses were slightly stasis and dilated, the hepatic cells were slightly edema and granular degeneration, and lymphocyte infiltration was observed. Immunohistochemical staining results showed that in the control group, a large number of positive expressions of brown insulin was observed. The cells were closely arranged and uniform with complete morphology. In the model group, the islets were less stained and irregular in shape. The average optical density of insulin in islets in model group was lower than that in control group, and the difference was statistically significant (P < 0.05). Conclusion Islet tissue damage, decreased insulin expression and elevated blood glucose occurred in early stage after severe heat stroke in pigs, which may be related to the impaired islet function.
