Abstract:Objective To investigate the correlation of peripheral blood microRNA (miR-155) expression with T helper (Th)1/Th17 cells and to related cytokines in children with atopic dermatitis (AD). Methods A total of 89 AD children admitted to Taihe Hospital, Hubei Province (hereinafter referred to as “our hospital”) from February 2018 to September 2021 were selected as AD group, and according to the score of scoring atopic dermatitis, they were divided into mild group (28 cases), moderate group (37 cases), and severe group ( 24 cases). Another 25 healthy children who underwent physical examination during the same period were selected as control group. The levels of miR-155 in peripheral blood, Th1/Th17 cells, and related cytokines of each group were compared, and the correlation between the expression of miR-155 in peripheral blood and Th1, Th17, and related cytokine levels in children with AD were analyzed. Results The expression of miR-155, the proportion of Th17 cell, interleukin (IL)-17, and IL-23 in AD group were higher than those in control group, and the proportion of Th1 cell, Th1/Th17, interferon-γ (IFN-γ), IL-2, and IFN-γ/IL-17 were lower than those in control group (P < 0.05). The expression of miR-155, the proportion of Th17 cell, IL-17, and IL-23 in severe group were higher than those in mild group and moderate group, while the proportion of Th1 cell, Th1/Th17, IFN-γ, IL-2, and IFN-γ/IL-17 were lower than those in mild group and moderate group (P < 0.05). The expression of miR-155, the proportion of Th17 cell, IL-17, and IL-23 in moderate group were higher than those in mild group, and the proportion of Th1 cell, Th1/Th17, IFN-γ, IL-2, and IFN-γ/IL-17 in moderate group were lower than those in mild group (P < 0.05). The expression of miR-155 in peripheral blood of AD children was positively correlated with the proportion of Th17 cell, serum IL-17 and IL-23 (r > 0, P < 0.05), and negatively correlated with the proportion of Th1 cell, Th1/Th17, IFN-γ, IL-2, and IFN-γ/IL-17 (r < 0, P < 0.05). Conclusion The expression of miR-155 in peripheral blood of AD children is increased, and it is related to the imbalance of Th1/Th17. The miR-155 may participate in the pathogenesis of AD by regulating Th1/Th17.
