Extraction and content determination of flavonoids from Mongolian Medicine Xanthoceras sorbifolia Bunge |

Abstract Objective To extract flavonoids from Xanthoceras sorbifolia Bunge and detect the contents of quercetin and epigallocatechin gallate (EGCG). Methods AB-8 macroporous resin was used for separation and elution with water ethanol system to concentrate the target compound. Chromatographic conditions: Hypersil C18 column (4.6 mm×200 mm,5 μm); flow rate 1.0 mL/min; column temperature 28℃, mobile phase acetonitrile-water, gradient elution; detection wavelength 225 nm; injection volume 20 μL. The content of quercetin and EGCG were determined. Results The linear values of quercetin and EGCG were 0.060-0.964 mg/mL and 0.062-0.984 mg/mL (r > 0.9990). The contents of quercetin and EGCG in Xanthoceras sorbifolia Bunge were 0.933, 0.865 mg, respectively. Conclusion The content of quercetin and EGCG are determined by HPLC with simple operation and good reproducibility, which provide an effective method for the quality control of Xanthoceras sorbifolia Bunge and provide a basis for the wide application of single drug active ingredient in clinical practice.
