Study on correlation of characteristic chromatograms of pieces, standard decoction, intermediate and formula granule of Lonicera japonica flos by HPLC

Abstract:Objective To study the correlation of characteristic chromatograms of pieces, standard decoction, intermediate and formula granule of Lonicera japonica flos. Methods The results were studied by HPLC method; column was Thermo Scientific AcclaimTM120 C18 (5 μm, 4.6×250 mm); acetonitrile -0.4% phosphoric acid aqueous solution was used as mobile phase for gradient elution. The detection wavelength was 327 nm. The column temperature was 30℃. The flow rate was 1 mL/min. The injection volume was 10 μL. Characteristic chromatograms and correlation with 15 batches of pieces, standard decoction, intermediate and formula granule of Lonicera japonica flos were compared. Results Six components, including neochlorogenic acid, chlorogenic acid, crypto-chlorogenic acid, isochlorogenic acid B, isochlorogenic acid A and isochlorogenic acid C, were identified by the characteristic chromatograms for 15 batches of pieces, standard decoction, intermediate and formula granule of Lonicera japonica flos. The similarity of characteristic chromatograms of 15 batches of pieces, standard decoction, intermediate and formula granule of Lonicera japonica flos were all greater than 0.9. The main chemical components of pieces, standard decoction, intermediate and formula granule of Lonicera japonica flos were basically the same. Conclusion The established HPLC characteristic chromatograms method can be used for the quality control of pieces, standard decoction, intermediate and formula granule of Lonicera japonica flos.
