Abstract:Objective To study the correction between transcription factors GATA-3,T-bet and TH cytokine in patients with cervical lesions and their expression level and significance in peripheral blood. Methods A total of 76 patients who were admitted to the First Affiliated Hospital of Xinjiang Medical University from January 2017 to January 2019 were selected. According to the cervical thinprep cytologic test, they were divided into control group (33 cases), low-grade squamous intraepithelial lesion (L-SIL) group (10 cases) and high-grade squamous intraepithelial lesion (H-SIL) group (33 cases). The expression levels of GATA-3 and T-bet in serum were detected by Rt-PCR, and the TH cytokine levels in serum of three groups were detected by flow cytometry. Pearson correlation analysis was used for correlation analysis. Results The expressions of interleukin (IL)-4 and IL-10 in H-SIL group and L-SIL group were higher than those in control group, and the expression of GATA-3 was higher than that in control group, while the expression of T-bet in L-SIL group was lower than that in control group, and the differences were statistically significant (all P < 0.05). IFN-γ was positively correlated with T-bet (r = 0.65, P < 0.05), and negatively correlated with GATA-3 (r = -0.51, P < 0.05). IL-4 was positively correlated with GATA -3 (r = 0.56, P < 0.05), and negatively correlated with T-bet (r = -0.72, P < 0.05). IL-10 was negatively correlated with T-bet (r = -0.65, P < 0.05). Conclusion Peripheral blood of patients with L-SIL shows dominance of TH1 cells and up-regulated T-bet gene expression, while that of patients of H-SIL shows dominance of TH2 cells and down-regulated T-bet gene expression. It can be seen that the occurrence of L-SIL and H-SIL is related to TH cytokines, GATA and T-bet.
