Effect of algae eucheuma striatum polysaccharide on mouse vascular smooth muscle cells injured by nano-hydroxyapatite crystals

Abstract:Objective To investigate the effect of algae eucheuma striatum polysaccharide (ESP) on mouse vascular smooth muscle cells injured (MOVAS) induced by nano-hydroxyapatite crystals (nano-HAP). Methods Cultured vascular smooth muscle cells were divided into the blank control group, the model group and the experimental group. Fresh medium was added to the control group, the model group was stimulated by nano-HAP for 24 h. The cells in the experimental group were treated with different concentrations (50, 150, 450 μg/mL) of ESP. The activity of cells was determined by MTT assay and LDH release by LDH kit, cell morphology was observed by hematoxylin-eosin staining. Results Compared with the blank control group, cell viability in the model group decreased significantly (P < 0.05), LDH release was significantly increased (P < 0.05). Compared with the model group, the activity of cells in the experimental group increased with the increase of ESP concentration, and the release of LDH decreased with the increase of ESP concentration, with statistically significant differences (all P < 0.05). Conclusion ESP can alleviate the damage of nano-HAP crystal to the membrane of vascular smooth muscle.
