Isolation and purification of verbascoside by macroporous resin

Abstract:Objective To study the purification process of the glycoside in Cistanche tubulosa (Schenk) Wight by macroporous resin. Methods The content of glycosides in Cistanche tubulosa (Schenk) Wight was determined by high performance liquid chromatography (HPLC). Three kinds of kinetic equations were applied to fit the experimental data. The isotherm adsorption lines at different temperatures were fitted by Freundlich and Langmuir equations, and the thermodynamic parameters were calculated. Finally, the purification of glycosides in Cistanche tubulosa (Schenk) Wight by macroporous resin was investigated by single factor test. Results D101 macroporous resin was selected. The purification conditions were as followed: the concentration of sample solution was 1.5 mg/mL, the maximum sample loading rate was 6 BV, and the sample speed was 2 BV/h. It was eluted with 11 BV purified water and 30% ethanol of 10 BV. The content of glucoside in the flower was 25.1% after purification by D101 resin. Conclusion Under the conditions of this process, the purification of glycosides in Cistanche tubulosa (Schenk) Wight by D101 macroporous resin is simple and stable, and it can increase the content of glycosides in Cistanche tubulosa (Schenk) Wight.
