斑蝥酸钠维生素B6对人成骨肉瘤细胞凋亡及存活素表达的影响

摘要目的研究斑蝥酸钠维生素B6诱导MG-63细胞凋亡的作用机制。方法应用不同浓度的斑蝥酸钠维生素B6作用于MG-63细胞，用MTT比色法检测药物对细胞的增殖抑制作用；反转录聚合酶链反应（RT-PCR）检测存活素信使核糖核酸的表达。结果斑蝥酸钠维生素B6在抑制MG-63细胞增殖方面有显著作用，其作用具有时间和剂量依赖性，抑制率可高达（68.56±5.21）%。RT-PCR显示可下调survivin基因表达。结论斑蝥酸钠维生素B6抑制MG-63细胞增殖效果明显，其机制可能与下调survivin基因的表达有关。

Abstract：Objective To study the mechanism of Sodium Cantharidate and vitamin B6 inducing apoptosis of MG-63 cells. Methods The different concentrations of Sodium Cantharidinate and vitamin B6 were applied in MG-63 cells, the anti-proliferative effect of drug for cells was detected by MTT colorimetry; reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of survivin messenger RNA. Results Sodium Cantharidate and vitamin B6 could inhibit the proliferation of MG-63 cells in a time and dose-dependent manner, and the inhibition rate could be as high as (68.56±5.21)%. RT-PCR showed downregulation of survivin gene expression. Conclusion The inhibitory effect of Sodium Cantharidate and vitamin B6 on the proliferation of MG-63 cells is obvious, and the mechanism may be related to down regulation of the expression of survivin.
