Application of RP-HPLC in determination of the three main kinds of components in the extract of Psoralea corylifolia

Abstract:Objective To determine psoralen, isopsoralen and bakuchiol from the extract of Psoralea corylifolia by RP-HPLC. Methods The contents of paoralen, isopsoralen and bakuchiol were determined on the Epic C18 (250 mm×4.6 mm, 5 μm) column, with methanol: water in gradient elution manner. The flow rate was 1.0 mL/min, the column temperature was maintained at 30℃, and the detection wavelength was set up at 250 nm. Results The linear ranges of paoralen, isopsoralen and bakuchiol were 0.0563-0.3379, 0.0523-0.3139 and 0.1280-0.7680 μg (r=0.9999). The average recovery of paoralen, isopsoralen and bakuchiol were 100.66%, 100.43% and 99.41%, RSD were 1.77%, 1.45% and 1.63% (n=6). Conclusion The method is convenient, accurate sensitive and repeatable, and can be used in the content control method of psoralen, isopsoralen and bakuchiol in extract of Psoralea corylifolia.
