Abstract:Objective To determine the expression levels of multifunction cytokine AK2 in kainic acid-induced epileptic rat, and to investigate whether AK2 participates in the pathogenesis of EP. Methods 60 male SD rats were divided into control group (10 cases) and model group (50 cases), the animal model of temporal lobe epilepsy was established in model group by intra cerebroventricular injection of KA. Then the rats in the model group were divided into 6 hours group, 12 hours group, 1 day group, 3 days group and 1 week group, with 10 cases in each group, according to the different time points after seizures. AK2 mRNA and protein expression in frontal lobe and hippocampus tissue were measured by semi-quantitative RT-PCR and Western blotting. Results In the model group, expression levels of AK2 in both front al lobe and hippocampus tissue were significant increased compared with the control group (P < 0.05). And the expression levels were reached the highest 3 days after seizures and then gradually declined 1 week later (P < 0.05). Conclusion AK2 may participate in the pathophysiological process of EP and provide a new mechanistic basis for the pathogenesis of epilepsy.
