Research on the effect of Tripterygium wilfordii attenuated toxicity by compatibility

Abstract:Objective To observe the toxicity effect of Tripterygium wilfordii and the influence on the toxicity after compatibility with Chinese nourishing kidney herbs. Methods Sixty SD rats were randomly divided into negative control group, Tripterygium wilfordii group and compatibility group (Tripterygium wilfordii combined with Cuscuta chinensis, Fructus Lycii and Epimedium davidii) according to the body weight of animals, with 20 rats in each group. All groups were administrated with corresponding drugs by intragastric administration once a day for 30 days, the rats of negative control group were treated with 0.5% sodium carboxyl methyl cellulose (CMC-Na). During the test, the physical signs, behaviors, gland secretion, respiratory and stool characters of animals were observed every day, and weight was measured every week. After 30 days, the animals were killed for the examination of hematology, hormone levels and pathology. Results Compared with the negative control group, the body weight of Tripterygium wilfordii group was grown slowly (P < 0.05), the hematological indexes such as white blood cell count (WBC), hemoglobin (HGB), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), reticulocyte count (RET) and RET% were decreased significantly (P < 0.05), neutrophile granulocyte (NEUT), NEUT% and the ratio of lymphocyte (LYMPH%) were decreased significantly (P < 0.05); hormone examination showed that follicle-stimulating hormone (FSH), luteinizing hormone (LH) and testosterone (TTE) were decreased (P < 0.05) in the Tripterygium wilfordii group; histopathologic examination showed that the male ductus epididymis was empty, the lumen was narrow, and no sperm or cell components in the lumen were found in the Tripterygium wilfordii group. Compared with Tripterygium wilfordii group, the body weight of compatibility group showed no significant difference, the hematology indexes such as WBC, HGB and RET were increased (P < 0.05), TTE level was increased (P < 0.05) and the degree of epididymal lesions was reduced in compatibility group. Conclusion Under the experimental conditions, Tripterygium wilfordii has obvious toxic effects on reproductive system and blood system. Compatibility of Cuscuta chinensis, Fructus Lycii and Epimedium davidii can reduce the toxicity of Tripterygium wilfordii on reproductive system and blood system, which has obvious effects of attenuated toxicity by compatibility.
