Effects of different acupuncture methods in Mongolian Medicine on monoamine neurotransmitters and PKA/CREB/BDNF pathway in prefrontal lobe of depressed rats

Abstract:Objective To observe the effects of different acupuncture methods on the neurotransmitter and protein kinase (PKA) activity, cyclophosphamide receptor binding protein (CREB) phosphorylation and brain-derived neurotrophic factor (BDNF) expression in the prefrontal cortex of depressed rats. To explore the antidepressant mechanism of different acupuncture methods in Mongolian medicine. Methods A total of 40 male Sprague-Dawley rats were divided into blank group, model group, Mongolian medical silver needle group, Mongolian medicine three balanced needle group and drug group according to random number table method, with 8 rats in each group. Rats in the other groups were orphaned except for the blank group and were given unpredictable 7 different stress stimulation for 28 days. Among them, 4 rats in the blank group were raised in each cage and fed with normal drinking water without any stimulation; the model group did not receive any treatment; the Mongolian medical silver needle group received the silver needle for stimulation of the Bada dry point, Heart acupuncture points and black and white points 1 h before the daily stress stimulation; acupuncture and acupuncture points of the Mongolian medicine three balanced needle group were the same as those in the Mongolian medical silver needle group, but one acupoint was selected daily for warm needle stimulation; the drug group was given 1 hour before stress stimulation daily, Fluoxetine hydrochloride was formulated into a 1 mg/1 mL solution in a 0.9% sodium chloride solution, and the drug group received intragastric administration according to a body weight of 2 mg/kg. After 28 days, the rats were decapitated and the prefrontal lobe was taken. The 5-hydroxytryptamine (5-HT), norepinephrine (NE), dopamine (DA) content, PKA activity, CREB phosphorylation and BDNF expression level were detected by enzyme-linked immunosorbent assay and Western blot. Results Compared with the blank group, the contents of 5-HT, NE and DA in the prefrontal cortex of the model group were significantly decreased after 28 days of modeling (P < 0.01), and the levels of PKA activity, CREB and BDNF expression were also significantly decreased (P < 0.05). Compared with the model group, the contents of NE, DA and 5-HT in the prefrontal cortex of Mongolian medicine three balanced needle group and drug group were significantly increased, the differences were statistically significant (P < 0.05), the contents of DA and 5-HT in the prefrontal cortex of Mongolian medical silver needle group were significantly increased, the differences were statistically significant (P < 0.05). Compared with the model group, the levels of PKA activity, CREB and BDNF expression in the prefrontal cortex of Mongolian medical silver needle group, Mongolian medicine three balanced needle group and drug group were significantly increased, the differences were statistically significant (P < 0.05). Conclusion Different needles of Mongolian medicine can effectively increase the content of 5-HT/NE/DA, the activity of PKA and the expression of CREB-BDNF, suggesting that different needles of Mongolian medicine have antidepressant effects.
