表皮生长因子协同体预处理对大鼠脑缺血再灌注的保护作用

摘要 目的 探讨表皮生长因子协同体(EGF-c)预处理对大鼠脑缺血再灌注(IR)的保护作用及其相关机制。 方法 将30只雄性SD大鼠按随机数字表法分为假手术组(sham组)、缺血再灌注组(IR组)及EGF-c预处理+IR组(EGF-c组)。采用线栓法制作大鼠大脑中动脉栓塞模型(MCAO)。术后24 h,应用2%氯代三苯基四氮唑(TTC)染色法检测脑梗死体积,酶联免疫吸附测定(ELISA)法检测脑组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,蛋白免疫印迹法(Western blot)检测高迁移率族蛋白1(HMGB1)的表达。 结果 与sham组比较,IR组脑梗死体积增大(P < 0.01),SOD活性降低(P < 0.01),MDA和HMGB1的含量增加(P < 0.01)。与IR组比较,EGF-c组脑梗死体积减小(P < 0.05),SOD活性上升(P < 0.01),MDA含量下降(P < 0.01),HMGB1的表达量下降(P < 0.05)。 结论 EGF-c预处理可能通过改善脑组织SOD活性、降低MDA含量、下调HMGB1的表达从而对大鼠IR起到保护作用。

Abstract :Objective To study the effect of epidermal growth factor-coordination (EGF-c) on cerebral ischemia reperfusion (IR) in rats as well as its mechanism. Methods Thirty male SD rats were divided into sham group, ischemia reperfusion group (IR group), EGF-c pretreatment and IR group (EGF-c group) by random number table method. The middle cerebral artery occlusion (MCAO) model of rats was made by suture method. 24 hours after operation, the cerebral infarction volume was detected by 2% triphenyl tetrazolium chloride (TTC) staining, the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in brain tissue were measured by enzyme-linked immunosorbent assay (ELISA), the expression of high mobility group protein 1 (HMGB1) was examined by Western blot. Results Compared with sham group, the infarction volume was increased (P < 0.01), the SOD activity was decreased (P < 0.01), the contents of MDA and HMGB1 were increased in IR group (P < 0.01). Compared with IR group, the infarction volume was decreased (P < 0.05), SOD activity was increased (P < 0.01), the content of MDA was decreased (P < 0.01), the expression of HMGB1 was decreased in EGF-c group (P < 0.05). Conclusion EGF-c pretreatment may play a protective effect on IR of rats by improving the SOD activity, decreasing the content of MDA and the expression of HMGB1.
