Abstract:Objective To explore the intervention effects and related mechanism of Tongxinluo on high glucose-induced human retinal capillary endothelial cells (HRCECs) injury. Methods The cells damaged model was established with high glucose. The HRCECs were divided into normal group, model group (30 mmol/L glucose), Tongxinluo low dose group (TXL-L group, 100 μg/mL Tongxinluo), Tongxinluo middle dose group (TXL-M group, 200 μg/mL Tongxinluo) and TXL high dose group (TXL-H group, 400 μg/mL Tongxinluo) by random number table method. The cell viability, contents of endothelin (ET)-1, interleukin (IL)-6 and intercellular adhesion molecule (ICAM)-1 in supernatant, and the endothelial nitric oxide synthase (eNOS) and nuclear factor (NF)-κB protein expression in cells of each group were detected. Results Compared with normal group, the cell viability was significantly decreased, the contents of ET-1, IL-6 and ICAM-1 in supernatant were significantly increased, the protein expression of eNOS was significantly down-regulated, and the protein expression of NF-κB was significantly up-regulated in model group (P < 0.01). Compared with model group, the contents of ET-1 in supernatant were significantly decreased in TXL-M group and TXL-H group (P < 0.01), and the contents of IL-6 and ICAM-1 in supernatant were significantly decreased, the protein expression of eNOS was significantly up-regulated, and the protein expression of NF-κB was significantly down-regulated in TXL-L group, TXL-M group and TXL-H group (P < 0.05 or P < 0.01). Conclusion Tongxinluo can signifcantly improve high glucose-induced HRCECs injury, and plays a role in cell protection through reducing inflammatory injury.
