Evaluation of diagnosis value of plasma miRNA-21 and miRNA-142-5p in renal cell carcinoma

Abstract:Objective To investigate the clinical values of plasma miRNA-21 and miRNA-142-5p expression levels in the diagnosis of renal cell carcinoma (RCC). Methods Blood samples of patients with RCC and bebign renal disease from July 2015 to October 2017 in the Second People′s Hospital of Huai′an were collected, there were 55 cases and 49 cases respectively. The expression levels of plasma miRNA-21 and miRNA-142-5p were determined by fluorescence quantitative PCR in these patients. The correlations between the two miRNAs levels in plasma and clinicopathological characteristics of patients with RCC were analyzed. The diagnostic performance of each miRNA to distinguish RCC from benign renal disease was evaluated by the receiver operating characteristic (ROC) curve analysis. Results The levels of plasma miRNA-21 and miRNA-142-5p in RCC patients were higher than those in benign renal disease (P < 0.01). There was no significant correlation between plasma miRNA-142-5p levels and clinicopathological characteristics of RCC patients (P > 0.05), whereas the expression levels of plasma miRNA-21 was correlated with clinical stages (P < 0.05). There was no significant correlation between the expression of miRNA-21 and miRNA-142-5p in the plasma of 55 patients with RCC (P > 0.05). The area under ROC curve of the two miRNAs were 0.826 (95%CI:0.794-0.912) and 0.805 (95%CI:0.744-0.919). The sensitivity and specificity were 81.8% and 83.7% for miRNA-21, and 80.0% and 77.6% for miRNA-142-5p, respectively, for diagnosing RCC. And the sensitivity was increased to 87.3% with miRNA-21 in combination with miRNA-142-5p, despite slightly decreased specificity which was 75.5%. Conclusion These findings demonstrated that plasma miRNA-21 and miRNA-142-5p may serve as a promising biomarker in the diagnosis of RCC.
