Investigation of microbial contamination and thermoduric bacteria in fermented traditional Chinese medicine decoction pieces contained Sojae Semen Praeparatum and Massa Medicata Fermentata

Abstract:Objective To investigate the microbial contamination and the number and types of thermoduric bacteria in fermented traditional Chinese medicine decoction pieces contained Sojae Semen Praeparatum and Massa Medicata Fermentata decoction pieces, and to provide the basis for the establishment of microbial limit standards applicable to fermented Chinese traditional medicine decoction pieces such as Sojae Semen Praeparatum decoction pieces and Massa Medicata Fermentata decoction pieces. Methods Ten batches of Sojae Semen Praeparatum decoction pieces and five batches of Massa Medicata Fermentata decoction pieces were collected. Aerobac teria, mold and yeasts and control bacteria were checked according to the 2015 edition of Chinese Pharmacopoia. After decocting at 100℃ for 30 min, the number of thermoduric bacteria was investigated, and the dominant thermoduric bacteria were identified by Biolog automatic microbial identification system. Results In different batches of Sojae Semen Praeparatum decoction pieces, quantities of aerobic bacteria ranged from 104-108 cfu/g and that of molds and yeasts from 103-108 cfu/g; in different batches of Massa Medicata Fermentata decoction pieces, quantities of aerobic bacteria, molds and yeasts all ranged from 103-105 cfu/g. All ten batches of Sojae Semen Praeparatum decoction pieces were found control bacteria, and four batches of Massa Medicata Fermentata decoction pieces were found control bacteria. The number of thermoduric bacteria was within 103-108 cfu/g and 102 cfu/g, respectively. All control bacteria were identified as Bacillus, and Bacillus subtilis accounted for a high proportion. Conclusion Traditional Chinese medicine decoction pieces fermented by natural fermentation are seriously polluted by internal and external microorganisms, so pure bacterial strains should be used for fermentation. Microorganisms of different fermented Chinese medicine decoction pieces have great differences, and corresponding microbial limit standards should be established for different varieties to ensure the safety and effectiveness of medicines.
