Study on the kidney-tonifying effect and mechanism of Shenbao Tablets

Abstract:Objective To study the kidney-tonifying effect of Shenbao Tablets and explore its mechanism. Methods The model of kidney-yang deficiency of Wistar rats induced by adenine was adopted. They were randomly divided into normal control group, model control group, Shenbao Tablets 1.14, 0.57 and 0.29 g/kg dose groups, Sildenafil Citrate Tablets group, with 13 rats in each group. Except the normal control group, the other rats were gavaged with adenosine 500 mg/kg for 10 d. On the second day of modeling, each group was administrated by gavage for 24 d. The body weight, the number of independent activities, the survival time of swimming at low temperature, the serum testosterone (T), blood urea nitrogen (BUN), creatinine (Cre), serum superoxide dismutase (SOD), malondialdehyde (MDA), cyclic guanosine phosphate (cGMP) and total nitric oxide synthase (NOS) were compared in each group. The castrated rat model was used, and 12 rats were randomly selected according to the body weight as the normal control group. The remaining rats were castrated. The castrated rats were randomly divided into model control group, Shenbao Tablets 2.28, 1.14 and 0.57 g/kg dose groups, and Sildenafil Citrate Tablets group, with 12 rats in each group. Each group was administrated by gavage once a day for 24 d, normal control group and model control group were gavaged the same volume of distilled water. The erectile latency,intracavernous pressure/blood pressure (ICP/BP), cGMP and total NOS levels and serum T levels were compared in each group. Results The body weight, the number of independent activities, the survival time of low temperature swimming, serum T levels and cGMP in penile sponge in model control group of kidney-yang deficiency were lower than those in normal control group, and serum BUN, Cre and MDA levels were higher than those in normal control group (P < 0.05). The survival time of low temperature swimming of Shenbao Tablets 1.14, 0.57 and 0.29 g/kg dose groups were longer than those of model control group, the BUN content was lower than that of model control group, the number of independent activities and cGMP levels in penile sponge of Shenbao Tablets 1.14 and 0.57 g/kg dose groups were higher than those of model control group, the serum T content and SOD activity of Shenbao Tablets 0.57 g/kg dose group were higher than those of model control group, and the Cre content was lower than that of model control group (all P < 0.05). There were no significant differences in the body weight, serum MDA content and total NOS levels in penile sponge in Shenbao Tablets 1.14, 0.57, 0.29 g/kg dose groups and model control group (P > 0.05). The erectile latency of castrated rats in model control group was higher than that in normal control group, and the serum T content, ICP value, ICP/BP ratio, cGMP level of penile sponge were lower than those in normal control group (P < 0.01). The erectile latency of Shenbao Tablets 2.28, 1.14 and 0.57 g/kg dose groups were lower than those of model control group, the serum T content, ICP value and ICP/BP ratio of penile sponge were higher than those of model control group, the cGMP levels of penile sponge in Shenbao Tablets 2.28 g/kg dose group were higher than those of model control group (all P < 0.05). There were no significant differences in BP value and total NOS of penile sponge in Shenbao Tablets 2.28, 1.14 and 0.57 g/kg dose groups and model control group (P > 0.05). Conclusion Shenbao Tablets has the effect of kidney-tonifying, and its mechanism is related to the increase of androgen content, the enhancement of NO/cGMP pathway activity and SOD activity.
