Effect of Bushen Huatan Decoction on bone remodeling balance in ovariectomized osteoporosis rats

Abstract:Objective To study the effect of Bushen Huatan Decoction on bone remodeling balance in ovariectomized osteoporosis rats. Methods Thirty-six month old female SD rats were divided into three groups: sham operation group, model group and traditional Chinese medicine group according to the random number table method, with 10 rats in each group. In sham operation group and traditional Chinese medicine group, the model was made by ovariectomy. In sham operation group, only the same amount of fat was removed. 5 weeks after operation, rats in traditional Chinese medicine group were given Bushen Huatan Decoction, and rats in sham operation group and model group were given the same amount of normal saline. 12 weeks after rats were killed, the content and localization of bone morphogenetic protein 2 (BMP2), runt related transcription factor 2 (RUNX2), osteoprotegerin (OPG), nuclear factor kappa B receptor activating factor ligand (RANKL) in serum and bone tissue were detected by enzyme-linked immunosorbent assay, Western blot and immunohistochemistry respectively. Bone mineral density of rats was measured by micro CT, and the differences of each index between each group were analyzed. Results Bone mineral density and serum BMP2, RUNX2, OPG and OPG/RANKL contents of model group and traditional Chinese medicine group were lower than those of sham operation group, and RANKL content was higher than that of sham operation group, with statistically significant differences (P < 0.05). Bone mineral density and serum BMP2, RUNX2, OPG, OPG/RANKL content of the rats in traditional Chinese medicine group were higher than those in model group, and RANKL content was lower than that in model group, with statistically significant difference (P < 0.05). BMP2, RUNX2, OPG and OPG/RANKL gray values of bone tissue in model group and traditional Chinese medicine group were lower than those in sham operation group, and the RANKL gray values were higher than those in sham operation group (P < 0.05). BMP2, RUNX2, OPG and OPG/RANKL gray values of bone tissue in traditional Chinese medicine group were higher than those in model group, and RANKL gray values were lower than those in model group, the differences were statistically significant (P < 0.05). The results of immunohistochemistry showed that the positive expression of BMP2, RUNX2 and OPG was mainly located in the cytoplasm of bone marrow stromal cells and osteoblasts around the trabecula; the positive expression of RANKL was located in osteoclasts at the border of bone, which were brownish yellow granules. Conclusion Bushen Huatan Decoction can regulate bone remodeling balance through BMP2/Smad/RUNX2 signal pathway and OPG/RANKL/RANK signal pathway to treat postmenopausal osteoporosis.
