Correlation study of expression of ADAM8, IRF1 in peripheral blood and pulmonary function in children with bronchial asthma

Abstract:Objective To study the correlation of the expression of deintegrin metalloproteinase 8 (ADAM8), interferon regulatory factor 1 (IRF1) in peripheral blood and pulmonary function in children with bronchial asthma (BA). Methods Eighty-four children with BA who were treated in Haikou Women & Children Hospital ("our hospital" for short) from January to August 2018 were selected as observation group, and they were divided into mild group (30 cases), moderate group (44 cases) and severe group (10 cases) according to the severity of the disease. Eighty healthy children who underwent physical examination in our hospital at the same time were selected as control group. The pulmonary function indices, the expression levels of ADAM8, IRF1 mRNA and protein in peripheral blood of each group were detected and compared. The correlation between the expression of ADAM8, IRF1 protein in peripheral blood and pulmonary function indices was analyzed. Results The expression levels of ADAM8, IRF1 mRNA and protein in peripheral blood of observation group and mild, moderate, severe group were significantly higher than those of the control group, the differences were highly statistically significant (P < 0.01); the expression levels of ADAM8, IRF1 mRNA and protein in peripheral blood of the severe group and moderate group were significantly higher than the mild group, and those of the severe group were significantly higher than the moderate group, the differences were statistically significant (P < 0.05). The forced expiratory volume in 1 second (FEV1), FEV1/forced vital capacity (FEV1/FVC), peak expiratory flow (PEF) in the observation group and mild, moderate, severe group were significantly lower than the control group, the differences were statistically significant (P < 0.05). The levels of FEV1, FEV1/FVC and PEF in the severe group and moderate group were significantly lower than the mild group, while the severe group was significantly lower than the moderate group, the differences were statistically significant (P < 0.05). Pearson correlation analysis showed that, the expression of ADAM8, IRF1 protein was negatively correlated with FEV1, FEV1/FVC and PEF (rADAM8=-0.780, -0.773, -0.682, P < 0.01; rIRF1=-0.684, -0.710, -0.768, P < 0.01). Conclusion The expression of levels ADAM8 and IRF1 in peripheral blood of children with BA are significantly increased, and it is closely related to pulmonary function. It can be considered as the monitoring target of children with BA, in order to assist the diagnosis and treatment process.
