Effect of Fibrauretin on the protein expression and activation of NF-κB in lung tissues of mice with acute lung injury

Abstract:Objective To study the effect of Fibrauretin on the protein expression and activation of nuclear factor kappa B (NF-κB) in lung tissues of mice with acute lung injury (ALI). Methods Sixty male SPF Kunming mice were randomly divided into 6 groups by drawing lots (10 mice in each group): ALI group [(intratracheal instillation of 5 mg/kg lipopolysaccharide (LPS)], normal control group (sham operation), positive control group (3.3 mg/kg Hydrocortisone Injection + LPS), low, medium and high doses of Fibrauretin groups (2, 10 mg/kg and 50 mg/kg Fibrauretin Injection + LPS). The positive drug and Fibrauretin were given by intraperitoneal injection once a day for 3 consecutive days before establishment of ALI model. Immunohistochemistry was used to detect the expression and localization of NF-κB p65 protein in lung tissues. Western blot was used to detect the expression of phosphorylated NF-κB p65 protein in lung tissues. Results Compared with ALI group, the percentage of positive cells with the entry of NF-κB p65 protein into the nucleus in lung tissues of mice in medium and high doses of Fibrauretin groups was decreased significantly, the difference was statistically significant (P < 0.05). Compared with ALI group, the protein expression of phosphorylated NF-κB p65 in lung tissues of mice in low, medium and high doses of Fibrauretin group was down-regulated significantly, the differences were statistically significant (P < 0.05 or P < 0.01). Conclusion Fibrauretin has preventive effect on LPS-induced acute lung injury in mice, and the effect is related to the inhibition of the protein expression and activation of NF-κB.
