Effects of bone mesenchymal cells transplantation on number of CD4+CD25+Treg cells of peripheral blood in atherosclerotic rats

Abstract:Objective To explore the effect on the number of CD4+CD25+regulatory T cells in the peripheral blood of the rats with AS after the mesenchymal stem cells transplanted into them and the effects of the two cells on the related inflammatory factors and AS. Methods 90 male Wistar rats were divideinto two groups: the normal diet group (30 rats) and the high-fat diet group (60 rats), and the high-fat diet group were divide into the control group (30 rats) and the transplanted group (30 rats). MSCS suspension (1×106 cells dissolved in 200 μL PBS) were injected into the transplanted group via the tail vein, while the control group were given 200 μL PBS. TC, TG, HDL-C, and LDL-C were analyze by automatic biochemical analyzer, the levels of hs-CRP and IL-6 in peripheral blood serum were measurated by ELISA and the level of CD4+CD25+Foxp3 Treg cells were observed by flow cytometry and the histological changes were observed by hematoxyli eosin stain. Results The histological analysis showed that the model of atherosclerosis was formed sucessfully. The blood lipid and inflammatory factors of control group were than the normal diet group(P < 0.05). The levels of blood lipid and inflammatory factors the transplanted group decreased after 4 weeks (P < 0.05), and the number of CD4+CD25+ regulatory T cells and CD4+CD25+Foxp3 increased(P < 0.05). Conclusion The transplantation of BMMSCs to Wistar rats with AS can increase the number of CD4+CD25+regulatory T cells, promote the expression of Foxp3, and decline blood lipid and inflammatory factors, thus providing a new idea to restrain the occurrence and development of AS and stabilize the plaques.
