Influences of Bushen Huoxue Granules on M1/M2 macrophage polarization in the heart of ovariectomized mice

Abstract:Objective To explore whether Bushen Huoxue Granules could cure cardiac microvascular dysfunction by regulating M1/M2 macrophage polarization in female ovariectomized mice. Methods According to random number table, 30 SPF level and 8 weeks old C57BL/6 mice were divided into sham group, model group and treatment group. Eight weeks after ovariectomy, treatment group was given Bushen Huoxue Granules (containing crude drugs 6 g/mL) by gavage, once a day, for eight weeks. Sham group and model group were given the same volume of physiological saline. HE staining, immunofluorescence staining and real-time quantitative PCR were used to detect the influences of Bushen Huoxue Granules on organization structures of heart tissue, the ultra oxygen anion, platelets endothelial cell adhesion molecules (CD31), CD68, and the mRNA expression of interleukin (IL)-1β, tumor necrosis factor (TNF)-α, IL-10 and vascular endothelial growth factor (VEGF). Results Compared with the sham group, the expression of IL-1β mRNA, TNF-α mRNA, ultra oxygen anion, CD68 cells in model group were increased (P < 0.05), while the expression of CD31, IL-10 mRNA were decreased (P < 0.05). Compared with the model group, the expression of IL-1β mRNA, TNF-α mRNA, ultra oxygen anion of heart tissue in the treatment group were decreased (P < 0.05), while the expression of CD31 and IL-10 mRNA were increased (P < 0.05). No significant difference was found with the expression of CD68 between the model group and treatment group (P > 0.05). Conclusion Bushen Huoxue Granules may cure cardiac microvascular dysfunction by regulating M1/M2 macrophage polarization in ovariectomized mice.
